The effects of ultrasonic-assisted ultrasonic time, concentrations of ethanol and ratio for material and liquid on the extraction yield of isoflavones were investigated for optimizing the ultrasonic-assisted extraction process of isoflavones from cicer arietinum L. On the basis of single factors, the best extraction process of isoflavones in Cicer arietinum L. was obtained by using the response surface combined with genetic algorithm: the extraction time of1.73 h, the ethanol concentration of 78.64% and the liquid ratio of 1:24.9 g/mL, and the extraction rate of isoflavones in cicer arietinum L. of 0.76 mg/g. As the 5-lipoxygenase was the target, the potential anti-inflammatory components with 5-lipoxygenase inhibition in cicer arietinum L. were screened by ultrafiltration mass spectrometry (UF-MS). The results indicated that the extraction rate of active ingredients in cicer arietinum L. could be further optimized by using the response surface binding genetic algorithm, and the potential anti-inflammatory active ingredients in cicer arietinum L. were daidzin, daidzein, calycosin, homoferreinrin and biochanin A, with degree of binding of 40.82%, 36.93%, 31.65%, 28.75% and 22.51%, respectively.
