摘要
目的:建立HPLC同时测定羚羊清肺颗粒中绿原酸、栀子苷、橙皮苷、黄芩苷和甘草酸含量的方法;采用Agilent TCC18色谱柱(4.6mm×150mm,5μm),流动相为乙腈(A)-0.6%磷酸水溶液(B),梯度洗脱程序为(020min,4%→10%A;2024min,10%→8%A;2425min,8%A→10%A;2528min,10%A→12%A;2830min,12%A→23%A;3040min,23%A→30%A;4045min,30%A→35%A;4546min,35%→40%A;4650min,40%→50%A;5055min,50%→60%A;),流速为1.0m L·min-1,柱温为30oC,检测波长分别为238nm(栀子苷),250nm(甘草酸),280nm(橙皮苷、黄芩苷),327nm(绿原酸)。结果:绿原酸、栀子苷、橙皮苷、黄芩苷和甘草酸进样量分别在0.04702.8217μg(r=0.9996),0.027081.6247μg(r=0.9996),0.00771.5474μg(r=0.9999),0.02351.4100μg(r=0.9999),0.00720.4333μg(r=0.9996)范围内线性关系良好;平均加样回收率(n=6)分别为96.93%、98.04%、97.10%、101.15%和95.93%,RSD分别为1.62%、1.15%、0.57%、0.80%和1.71%。结论:本法适用于同时测定羚羊清肺颗粒中绿原酸、栀子苷、橙皮苷、黄芩苷和甘草酸的含量。
