1型糖尿病小鼠心房肌细胞电重塑及AGE对其的影响

摘要

Aim To explore type 1 diabetes mice and the advance glycation end products (AGE) involved in electrical remodeling of atrial myocytes. Methods The diabetic mouse model was induced by intraperitoneal injection of STZ; action potential duration, and the current density of ICa,L, Ito and Ikur were detected by whole-cell patch technique in atrial myocytes of diabetic mice; the expression of AGE, RAGE and ion channel proteins in the atrial tissues of mice was measured by Western blot; HL-1 cells were treated with AGE/RAGE to detect the expression of ion channel proteins. Results The level of random blood glucose was markedly higher than that in control group, the action potential duration was significantly prolonged and the current density of ICa,L, Ito Ikur and the expression of channel proteins Cav1.2, Kv4. 3 and Kv1.5 markedly decreased in atrial myocytes of diabetic mice, while AGE and RAGE proteins increased. Conclusions The action potential of atrial myocytes is significantly prolonged in type 1 diabetic mice, while the current density of ICa,L, Ito, Ikur and the expression of their channel proteins decrease; AGE is involved in the electrical remodeling of HL-1 atrial myocytes. ? 2021 Publication Centre of Anhui Medical University.
