OBJECTIVE: As a member of the kinesin superfamily,KIF23 was involved in cell division,chromosome movement,and cell trafficking.In recent years,more and more attention had been paid to its role in the occurrence,progress and prognosis of malignant tumors,and it was expected to become a new target for cancer treatment.This study was to investigate the effect of silencing KIF23 gene on proliferation,migration and invasiveness of epithelial ovarian cancer cells.METHODS: The epithelial ovarian cancer SKOV3 cells were cultured and divided into siRNA-KIF23 group,negative control group and blank group.Quantitative real-time polymerase chain reaction(qRT-PCR) was used to detect the expression of KIF23 gene,MTT and cell cloning assay were used to detect cell proliferation,scratch test and Transwell method were used to detect cell migration and invasion ability.RESULTS: The relative expression of KIF23 mRNA in the siRNA-KIF23 group was 0.24±0.10,the negative control group was 1.02±0.07,and the blank group was 1.01±0.05. The difference between the three groups was statistically significant,F=217.090,P<0.001.Compared on the absorbance A values at 24,48,72 and 96 h among the siRNA-KIF23 group,negative control group and blank group,the difference was statistically significant,Fgroup=44.187,Pgroup<0.001.The number of cell clones in the siRNA-KIF23 group was 28.94±2.07,which was lower than that in the negative control group (58.43±5.64) and blank group (54.48±4.14),the difference was statistically significant,F=86.656,P<0.001.At 24 and 48 h,the scratch healing rate was (7.16±1.38)% and (30.85±6.63)% in the siRNA-KIF23 group,(51.38±6.52)% and (76.68±7.04)% in the negative control group,and (48.18±4.44)% and (75.41±5.25)% in the blank group,respectively,and the difference were statistically significant,Fgroup=592.715,Pgroup<0.001.The number of invasive cells in the siRNA-KIF23 group was 87.94±5.96,which was lower than that in the negative control group (121.32±7.73) and blank group (117.23±7.09),and the difference was statistically significant,F=41.002,P<0.001.CONCLUSON Silencing the expression of KIF23 gene can reduce cell proliferation and inhibit cell migration and invasion.
