Flavonoids'biological activity can often be improved by coordinating flavonoids with metal ions.The coordination ratio of neohesperidin dihydrochalcone(NHDC) to Cu2+was studied by the method of mole ratios and the method of continuous variations of equimolar solutions.The structure of NHDC before and after the coordination was studied by UV, IR and XRD analysis.DPPH method, salicylic acid method and FRAP method were used to compare the antioxidant activity of NHDC and its complex. Results indicate that the coordination ratio of NHDC to copper is 2:1;the stability of the NHDC-Cu (Ⅱ) complex is greatly affected by pH, and the stablest state is at pH7.0. The coordination can significantly enhance the hydroxyl radical-scavenging activity of NHDC. The reason may be that the coordination with copper ions improves the water solubility and increases the amorphous degree of NHDC. And the increase of water solubility and bioavailability after the coordination may be attributed to the "concentrated effect"of copper ions on the non-sugar ligand part of NHDC and the"embedding effect"of the sugar part of NHDC on the non-sugar part after the coordination.However, NHDC or its complex with Cu2+has no significant difference for their scavenging ability on fat-soluble free radicals like DPPH? and for their total reduction force.
