  The content of acetate in the liquor of pretreated biomass is potential corresponding to the degradation of hemicellulose in biomass.It is significant to determine the acetate acid in the pretreatment liquor.A facile method was established in this paper to determine the content of acetic acid in bamboo hydrolysate by high performance liquid chromatography with a Zorbax SB-C18 column eluted with the buffer of NaH2PO4 solution in 0.01 mol/L (pH2.98) and methanol (98 ∶ 2) in a flow rate of 1 ml/min under 25℃, and detected at wavelength of 210nm.The result showed that there was a well linear relationship between the peak area and the concentration of acetic acid at range of acetic acid: 0.01-1 g/L(r =0.9999) ; the average recovery of acetate acid was 99.74% and RSD was1.92%.In summary, the facile and accurate method is suitable for determination of acetic acid in bamboo pretreatment hydrolysate.
