摘要

目的考察甘草对RSV、HSV-1、EV71、CV-B3、CV-B5的抑制作用,并筛选其抗RSV活性部位,研究其体内外抗RSV机制。方法采用CPE法结合MTT法,以TI值为指标考察甘草抗病毒作用,体外加时实验考察其有效部位对RSV复制周期的影响。建立RSV感染小鼠模型,于给药第5天解剖,考察肺指数变化,HE染色观察肺组织病变情况,ELISA法检测血清炎症、免疫、氧化应激相关因子水平。结果甘草水提物对5种病毒均有一定的抑制作用,TI值分别为42.00、19.23、22.73、40.50、28.00,对RSV作用最强。甘草抗RSV活性部位为正丁醇及剩余水部位,TI值分别为36.22、>15.71,并作用于RSV复制周期的吸附、穿入、复制阶段,其中正丁醇部位可改善RSV所带来的肺组织损伤,降低感染小鼠肺指数,能降低NF-κB、TLR4、TNF-α、keap1水平,提高Nrf2、IFN-β水平。结论甘草对RSV、HSV-1、EV71、CV-B3、CV-B5均具有一定的抑制作用,其正丁醇部位可通过抑杀病毒、降低氧化应激水平、调节免疫、控制炎症来改善RSV引起的肺组织损伤。
