谷 proposta uma nova metodologia para inocula o de Uromyces appendiculatus a fim de avaliar a resist那ncia do feijoeiro 角 ferrugem. As linhagens Golden Gate Wax (gene Ur-6) e Ouro Negro (gene Ur-ON) foram cruzadas para a obten o de uma popula o F2, que foiinoculada com a ra a 29-3 de U. appendiculatus. Essa ra a 谷 incompat赤vel com Ouro Negro e compat赤vel com Golden Gate Wax. Folhas prim芍rias das plantas F2 foram destacadas dez dias ap車s a semeadura e inoculadas por submers o em suspens o de in車culo. Posteriormente, cada folha foi transferida para uma placa de Petri contendo papel de filtro umedecido com 芍gua destilada, e as placas foram, ent o, incubadas em BOD a 20~C, em regime de 12h de luz dia-1. A umidade nointerior das placas foi mantida pela adi o de 1,5 mL de 芍gua destilada em intervalos de tr那s dias. Como controle, as folhas prim芍rias remanescentes das plantas F2, no mesmo est芍gio dedesenvolvimento, foram inoculadas utilizando o m谷todo convencional de inocula o. Ambos os procedimentos de inocula o foram eficientes para avaliar a rea o do feijoeiro ao U. appendiculatus. Em ambos os m谷todos, a an芍lise fenot赤pica da popula o F2 confirmou anatureza monog那nica dominante da resist那ncia 角 ferrugem conferida pelo gene Ur-ON. A new methodology of inoculating Uromyces appendiculatusin tests aimed at evaluation of common bean resistance to rust is proposed. The common bean lines Golden Gate Wax (gene Ur-6) and Ouro Negro (gene Ur-ON) were crossed to obtain a F2 population, which was inoculated with race 29-3 of U. appendiculatus. Race 29-3 is incompatible with &Ouro Negro* and compatible with &Golden Gate Wax*. Primary leaves of ten-days-old F2 plants were detached and inoculated by immersion into an inoculum suspension. Immediatelyfollowing immersion, each F2 leaf was placed in a Petri dish containing a filter paper previously moistened with distilled water. These dishes were incubated in a BOD at 20~C, under a 12hdaily light regime. The humidity in the dishes was maintained by addiction of 1.5 mL of distilled water in filter paper, under a three days regime. As control, the remaining primary leaves of tendays-old F2 plants were inoculated using the conventional inoculation method. Both inoculation methods were efficient in evaluating the reaction of the common bean to U. appendiculatus. Theresistance/susceptibility screening of the F2 population confirmed the monogenic dominant nature of the rust resistance controlled by the Ur-ON gene in both methods.
