This experiment was conducted to investigate the effects of dietary Macleaya cordata water extract on growth performance, diarrhea index and intestine health of weaned piglets challenged with enterotoxigenic Escherichia coli (ETEC) . Thirty 28-day-old healthy " Duroc×Landrace×Yorkshire" crossbred weaned piglets with body weight of (8.22±0.98) kg were randomly divided into 3 groups with 10 replicates per group and 1 pig per replicate. Piglets in the control group and model group were fed a basal diet, and those in the experimental group were fed the basal diet supplemented with 1 mL / kg Macleaya cordata water extract. The pre-trial period lasted for 3 days and the experimental period lasted for 18 days. On the 15th day of the experimental period, each pig in the model group and experimental group was given 10 mL of ETEC K88 bacterial solution at a concentration of 2×109 CFU / mL by gavage, while that in the control group was given the same dose of sterile LB culture medium by gavage. The results showed as follows: 1) from days 1 to 14 (before challenge), compared with the control group, the average daily gain of weaned piglets in the experimental group was significantly increased (P < 0. 05), and the ratio of feed to gain was significantly decreased (P < 0. 05) . There were no significant differences in the growth performance of weaned piglets among the groups on days 15 to 18 (after challenge) and the whole experimental period (days 1 to 18) (P>0.05) . 2) At 24 h after challenge, compared with the control group, the diarrhea index of weaned piglets in the model group was significantly increased (P<0.05); compared with the model group, the diarrhea index in the experimental group was slightly decreased, but the difference was not significant (P>0.05) . At 48 h after challenge, compared with the control group, the diarrhea index of weaned piglets in the model group was significantly increased (P < 0. 05); compared with the model group, the diarrhea index in the experimental group was significantly decreased (P< 0.05) . 3) Compared with the control group, the crypt depth in jejunum of weaned piglets in the model group was significantly increased (P< 0.05), and the ratio of villus height to crypt depth in jejunum was significantly decreased (P<0.05); compared with the model group, the ratio of villus height to crypt depth in jejunum as well as the villus height and ratio of villus height to crypt depth in ileum of weaned piglets in the experimental group were significantly increased (P<0.05), and the crypt depth in jejunum was significantly decreased (P< 0.05) . 4) Compared with the control group, the mRNA relative expression levels of interleukin-1β (IL-1β) in jejunum and interleukin-17 (IL-17), IL-1β, tumor necrosis factor-α (TNF-α) and interleukin-8 (IL-8) in ileum of weaned piglets in the model group were significantly increased (P < 0. 05); compared with the model group, the mRNA relative expression levels of interleukin-18 (IL-18), IL-8 and IL-1β in jejunum and IL-17, IL-1β, TNF-α and IL-8 in ileum of weaned piglets in the experimental group were significantly decreased (P< 0.05) . In conclusion, dietary 1 mL / kg Macleaya cordata water extract can improve the growth performance of weaned piglets, alleviate the diarrhea, growth performance decline, intestinal villus height decrease, crypt depth increase and intestinal inflammation caused by Escherichia coli infection.
